The analysis of antigen-experienced cells can be performed in vivo by using a unique model of experimental systemic lupus erythematosus (SLE) in the well-characterized, organ-specific, model of lupus, MRL-lpr/lpr mice. In the present study, we used the B cell-specific monoclonal antibody, 3.2.3, which recognizes a B cell activation antigen termed CD40 ligand (CD40L), to determine the impact of environmental stimuli on the clonally expanded memory B cells in this model. 3.2.3 preferentially stained B cells in SLE-affected kidneys, but not in the inflamed kidneys of non-lupus mice. Binding of the Ag was to >80% of B cells, and was inhibited by the Ab on MRL-lpr/lpr mice. Addition of the Ab to the inflamed lupus kidneys did not exacerbate the disease or the swelling of these lesions. The Ab also stained cells in the draining lymph nodes of the immunized mice that were in the transition between the naive and Ag-experienced B cells. However, draining lymph node B cells from a naive mouse did not stain for CD40L. To determine the IgG subclasses of the Ab reactive Ig, we used Fc-specific Ab to isolate the IgG subclass after protein G purification. The IgG1 subclass was the predominant subclass in the inflamed and draining lymph nodes, but IgG2a was produced preferentially in the lupus kidney.
